Summary
Progress in bioengineering, biotechnology and biomedicine is limited by our understanding of how genetic control systems work in living cells. The lack of methods for studying kinetics and gene regulation in single cells seriously impairs our prospects to gain deeper insight and to develop better quantitative models of such control systems. This proposal is divided into two main projects: The first project proposes two new single molecule techniques for studying kinetics in single cells at unprecedented time resolution. The first technique will make it possible to monitor coordinated regulation of different genes. The second technique will enable determination of dissociation rates between proteins in living cells. The second project will elucidate the importance of spatial and stochastic aspects of intracellular signal transduction by a combination of theoretical, computational and experimental approaches. The combination of state of the art single molecule microscopy in living cells with spatially extended stochastic modeling will significantly advance Sweden’s position in the field of systems biology. Both academic and industrial research will benefit greatly from the new methods to monitor and analyze intracellular control systems at high time resolution and single molecule sensitivity.
