Summary

Ageing is by far the biggest risk factor for a wide range of prevalent clinical conditions caused by abnormal accumulation of damaged protein aggregates. In this proposal, we will develop dynamic and stochastic mathematical models, laying the groundwork for novel strategies and deeper understanding of aging and age-related disorders. Our novel framework combines large scale kinetic model of the single cell and Bayesian nonlinear mixed effect model to better understand the dynamics governing the behavior of heterogeneous populations, and how single cells and cell-to-cell variability contribute to population fate. A stochastic model will be developed to assess the nature of aggregate inheritance. A web-based platform will be established that allows effective use of developed models through model simulator and visualization tool for descriptive representation and interpretation of the results. Predictive models developed within this project will generate novel insights into the understanding of age-related disorders, especially those associated to aggregate accumulation like Alzheimer’s disease and propose the best evolutionary strategies to achieve healthier population by eliminating accumulated aggregates thus delaying the onset of age-related diseases.

Åldrande är en process som förundrat människor sedan urminnes tider. Målet med forskning kring åldrande är att ge gamla människor ett friskare liv. Att ha en generation som kan njuta av livet som gammal, utan att vara en börda för samhället. Redan idag finns det indikationer på att vi inte bara lever längre, utan att vi dessutom mår bättre längre – någonting hälsoexperter refererar till som ”kompression av morbiditet”, vilket innebär att de flesta av oss bara kommer att lida svåra åldersrelaterade sjukdomar under de sista åren i livet. Vanlig bakjäst har accepterats som en mycket kraftfull modellorganism för att studera flera biologiska processer, inklusive åldrande. Den representerar en utmärkt modellorganism eftersom många grundläggande cellulära processer som förekommer i människor är bevarade i jäst. Allteftersom cellen åldras, ackumuleras proteinaggregat som visats vara en orsakande faktor i många åldersrelaterade sjukdomar såsom Alzheimers. Osjälvisk förmågan hos modercellen att hålla ackumulerade skador säkerställer att producerade dotterceller föds utan skador. Den exakta mekanismen för att producerar friska avkommor är okänd. Vi kombinerar matematisk modellering och experimentellt arbete för att förklara denna mekanism som kan få stor betydelse för att diagnostisera och behandla åldersrelaterade sjukdomar.
